AS2863619 is a potent, orally bioavailable inhibitor of cyclin-dependent kinases CDK8 and CDK19, exhibiting IC50 values of 0.61 nM and 4.28 nM, respectively. Pharmacological inhibition of CDK8/19 by AS2863619 enhances STAT5 activation, leading to induction of Foxp3 gene expression. AS2863619 facilitates the conversion of antigen-specific effector/memory T cells into Foxp3⁺ regulatory T (Treg) cells.
