At Ace Therapeutics, we develop humanized mice that express human cytokines using gene-targeting strategies, including IL-17 and IL-23. In these models, the murine exons encoding the extracellular domain are replaced with their human counterparts, ensuring expression of human cytokine proteins for psoriasis research. We use ELISA and qPCR to quantify the expression levels of human IL-17A and IL-17F. We also apply immunohistochemistry to profile T cell subsets, such as Th17, Th1, and Treg populations.
Our experts induce psoriasis lesions in humanized mice by topical application of imiquimod (IMQ). This stimulation drives strong activation of the IL-23/IL-17 axis, resulting in psoriasis phenotypes, including scaling, erythema, and epidermal thickening.
