Overview of Retinal Degeneration

Retinal degeneration describes a group of disorders that share progressive damage and loss of the neurovascular retina. Major categories include photoreceptor and retinal pigment epithelium (RPE) dystrophies, retinal ganglion cell (RGC) and optic nerve degeneration, and neurovascular injury such as diabetic retinopathy (DR) and other ischemic retinopathies. Different triggers, such as genetic defects, metabolic stress, ischemia, or toxic exposure, activate similar pathogenic cascades. Oxidative and metabolic stress, mitochondrial dysfunction, protein misfolding, excitotoxicity, and chronic inflammation injure neurons, glia, and vascular cells, disturb retinal layering and synaptic function, and finally impair visual signal processing.
